Runes are the persistent locks between runs in Dead Cells. Here's how to unlock them all starting from a fresh file.

1. Prisoner's Quarters -- Promenade of the Condemned -- VINE RUNE
2. Continue to Ramparts -- CUSTOM RUNE
3. Continue to Black Bridge -- VINE RUNE
**Optional reset (or ... once you die)
4. Prisoner's Quarters -- Toxic Sewers -- TELEPORTATION RUNE
**Optional reset (or ... once you die again)
5. Prisoner's Quarters -- Promenade of the Condemned -- Ossuary -- RAM RUNE
**Optional reset (again)
6. Prisoner's Quarters -- Toxic Sewers -- Ancient Sewers -- Insufferable Crypt -- Slumbering Sanctuary -- SPIDER RUNE
7. Continue to Forgotten Sepulcher -- EXPLORATION RUNE (see: Lag Rune)
8. Continue to Clock Tower -- High Peak Castle -- Throne Room -- HOMUNCULUS RUNE
