While the locations of these five "Harlequins" is available also in the main game, you need the DLC to find and beat up these guys.
The Harlequins are only aggressive regular characters which you need to beat up with fists only, and you don't want to kill them.

If you kill one by mistake, you can respawn him by using a "Tunnel" (move anywhere else, then return to where the Harlequin is) and then try again in the same position where you first found him (which will be the same of the video of course). Most of the times, you won't find him again at first, but it will be enough to run away for a bit and eventually he will be respawned in the exact same position where you first found him.

This procedure is also necessary if you trigger a fight with one of them, then flee without killing him, and eventually outrun him: he may not be back in his default position right as soon as you return there. If that's the case, as already said, just walk around (no need to go to the opposite side of the map, just sprint like past 3-4 buildings, for 100 meters or whatever you feel like) and return where the Harlequin was/will eventually be again.
