Note that due to the delayed departure from Sheffield this service operated non stop through Meadowhall, Chapeltown, Elsecar and Wombwell with the first stop being Barnsley where the service continued to call at all stations on time, this resulted in an end to end Sheffield to Huddersfiled time being around 8 minutes faster than scheduled.

Services on this route usually operate hourly calling at all stations. Additional Northern services operate between Sheffield and Barnsley either all stations or calling only at Meadowhall as part of the Nottingham/Sheffield to Leeds via Wakefield Kirkgate services. Other services operate between Sheffield and Meadowhall operated by Northern and TransPennine Express.

There is interchange at Sheffield with other Northern, TransPennine, East Midlands Railway and Cross Country services on the Midland Main Line. At Huddersfield there is TransPennine interchange with the North TransPennine route to Manchester/Liverpool and Leeds/York. At Meadowhall there is interchange with the Yellow route on the Sheffield Supertram tram network to Middlewood via Cathedral.

The rolling stock on this route is mainly 2 carriage class 150 Sprinter trains but 3 carriages are sometimes operated. Newer Northern trains such as the class 195 Civity do not operate on the Penistone line services.

This route is unelectrified throughout. This route is double track between Sheffield and Barnsley and single track for most of the route from Barnsley to Huddersfield with the exception of Shepley to Stocksmoor and at Penistone station. At Huddersfield Penistone line services use a bay platform to avoid crossing TransPennine services.
