The last scene is how you do the glitch. Just stand in front of the glowing door (or sword), quit to the title screen, and then continue or load any save. You'll be able to pray anywhere but you'll be locked into the prayer position and you'll be forced to close the game after. The only exception is if you pray near the door, as it'll open and Wander will stand back up. Same thing probably happens with the sword but I didn't check that.

You can actually open the door from very far away with this glitch. I've opened it from inside the Shrine of Worship and also from halfway across the field going toward Quadratus. Also, the one scene where the room wasn't loaded was actually on a save with 0 relics collected.

The glitch persists even if you pray at a statue or a corpse. So, you can try out the glitch with all of the colossi w/o having to go to them normally. You can seemingly do everything as normal while the glitch is active until you press the pray button.

I tried praying as shadow Wander but it didn't seem to work. I may have just messed up and forgot to go to the door first.

Note: The glitch doesn't seem to trigger with save shrines, corpses, or statues. Just the door and sword make the glitch happen. I tried it on the PS3 version but as expected, it doesn't work with anything.
