Leaf of Adhatoda vasica ( Vasaka ) is an important drug of Ayurveda, prescribed as an expectorant. For pharmacognosy and botany students it is very important to know the procedure for section cutting off any part of plant. To make a microscopic study we need to make a clear section of Adhatoda vasica ( Vasaka ). For the preparation of the section, we have followed following steps-

1- Collect fresh and clean leaves of Adhatoda vasica ( Vasaka )
2- Collect 04 watch glass which will be used in keeping the leaves of Adhatoda vasica ( Vasaka )
3- Place water, Safranin, Potato cube and again plain water in respective watch glass for Adhatoda vasica ( Vasaka )
4- Take a fresh leaf of Adhatoda vasica ( Vasaka ) and place it over the plain surface
5- now take a new and sharp razor blade to cut the leaf of Adhatoda vasica ( Vasaka )
6- Prepare the cubes of potato for further use for holding of Adhatoda vasica ( Vasaka )
7- Now cut the leaf of Adhatoda vasica ( Vasaka ) containing its midribs portion.
8- Transfer the cutted portion of Adhatoda vasica ( Vasaka ) in watch glass containing water
9- Now place that cut leaf of Adhatoda vasica ( Vasaka ) in between the opening of potato cubes.
10- Now cut the excess portion of leaves of Adhatoda vasica ( Vasaka ) through a razor blade.
11- Now move the blade through and forth direction
12- Place the leave of Adhatoda vasica ( Vasaka ) in watch glass containing water
13- Transfer the section of Adhatoda vasica ( Vasaka ) into a safranin contain watch glass
14- Now place that stained leaf of Adhatoda vasica ( Vasaka ) into watch glass containing plain water to remove excess color.
15- Place back the leaf into glass slide and add 2-3 drop of glycerin-water to prevent drying of Adhatoda vasica ( Vasaka )
16- Now add a coverslip over a slide containing leaf of Adhatoda vasica ( Vasaka )
17- Watch the leaf under a microscope and draw the structure.
