This video discusses an approach for non-polar extraction Solid Phase Extraction (SPE), including tips and tricks to boost productivity. The focus is on non-polar extraction SPE where the molecule with non-polar functional groups is extracted from predominantly polar matrices. The three main ways to eliminate unwanted contaminant are discussed. The first way is to leave the compound unretained (leaving it in the aqueous solution). Second, we can eliminate the unwanted contaminant during one or more wash steps. And finally, we can leave them bound to the sorbent when we elute our target compounds. There are also two simple approaches to method development for non-polar extractions: the first is mass balance and the second is the ten-bottle optimization. The mass balance approach is helpful for troubleshooting or optimizing an existing method. In this approach you monitor the behavior of your compound(s) of interest at each step by collecting the effluent after the cartridge is loaded, washed, and eluted. The ten-bottle optimization method helps to optimize the wash step to improve interference removal, while ensuring your target compounds are not removed. Agilent's Vac Elut SPS 24 vacuum manifold is also discussed.
