Gateway Cloning is a popular recombination based cloning method. It does not rely on restriction enzymes but instead uses integrases to recombine att sites between a final and donor vector. This causes an exchange of DNA segments that inserts the desired DNA into the final vector. There are two ways to achieve this: either with a BP reaction or with an LR reaction. In addition, gateway can be performed simply with typical attB/attP sites or with complex variants in a multisite reaction.
